Default USB help!

My computer isn't recognising my memory stick when I put it in the USB point. The little red light on the memory stick flashes like it's been recognised and the computer makes the pingping noise, but the E drive does not come up in My Computer. Also when I try to open documents saved on it, they don't open and say that the shortcut is unavailable.
I have my dissertation saved on my memory stick, so I really need some help to recover the files without formatting it!
